建立了对细胞摄入的铜纳米粒子(GuNPs)进行定量分析的荧光成像法。以近红外发光的谷胱甘肽-铜纳米粒子(GSH-CuNPs)与MCF-7细胞孵育,利用共聚焦显微镜在激发波长559 nm、发射波长600~700 nm条件下对MCF-7细胞内的GSH-CuNPs进行荧光成像。细胞经裂解、离心超滤后,收集GSH-CuNPs,采用电感耦合等离子体质谱(ICP-MS)对铜元素进行定量分析。通过SPSS统计分析关联胞内平均荧光强度与ICP-MS的定量检测数据,建立平均荧光强度与细胞中CuNPs的定量关系;再以ICP-MS测定细胞内铜元素的总含量,可获得由GSH-CuNPs转化为其它形式的铜含量。采用本方法研究了MCF-7细胞中GSH-CuNPs的代谢/转化过程。结果表明,GSH-CuNPs与细胞孵育50 min后,细胞内CuNPs含量由172.74 fg/cell降低至18.66 fg/cell,且GSH-CuNPs主要代谢转化为可溶性铜。
